Abstract
Colorectal cancer is an important contributor to mortality and morbidity
disease with highly heterogeneous genomic and transcriptomic levels. Nextgeneration sequencing (NGS) studies are becoming increasingly important tools for
exploring and detecting the role of genetic variation in complex diseases, including
cancer. Tissue and blood derived DNA are the current standard in whole-genome
sequencing (WGS) and whole-exome sequencing (WGS) as long as the quality and
quantity of human DNA in samples are sufficient. Recently, colorectal cancer is
characterized into distinctive four consensus molecular subtypes (CMS). These
subtypes provide a new paradigm for stratified treatment and drug repurposing. In
order to develop personalized medicine and to gain insight into the molecular wiring
and origin of CMS subtypes, reliable models are necessary. At here, Siriraj Hospital,
we have kept the buffy coats and cancer tissues from colorectal cancer patients in
Siriraj biobank. We were the first study to investigated and established the genome
landscape and database for Thai colorectal cancer from 130 patients. All genome
landscape from Thai colorectal cancer patients will provide and motivate the
innovation of cancer precision medicine in Thailand.
Recently, the consensus molecular subtype (CMS) in non-metastatic
colorectal cancer has clinical relevance, however the representativeness for
metastatic colorectal cancer is not well known. Here, we investigated the
heterogeneity metastatic CMS colorectal cancer using single cell spatial image
platform in combination with spatial transcriptomics, which may help to identify
and inform the development of metastasis-targeted therapies. The bulk whole
transcriptomic profiles from mRNA sequencing 30 metastatic primary colorectal
cancer were studied. Our platform will continually develop for clinical service.
Moreover, Thai genome database will initiate the frontier research in the future.
